SUPERIOR ACCURACY AND REPRODUCIBILITY
GSD-501 COMPARED TO FLUORESCENCE DETECTION FOR TWO-COLOR RECIPROCAL ASSAY
RLS was compared with direct fluorescent microarray detection in a
reciprocal two-color experiment, as shown in Figure 1. Spiked transcripts at
2x, 5x, and 10x relative to a 1 million copy control target were used to gauge
the differential output and sensitivity of the assay for both RLS and
fluorescence detection.
RLS Outperformed Fluorescence As Follows:
Both RLS channels effectively detected using 2x, 5x, and 10x with the
GSD-501
Cy5 detection could not be resolved at 1 million copies of control target
RLS two-color performance is based on approximately 7x more positives
than Cy5/Cy3
Reproducibility of RLS signals across the dynamic range of
the assay, as measured on 6 different GSD-501 instruments.

The experiment maintained a 1 million copy control level for Gold detection
while titrating Silver detection to 2x, 5x, and 10x copies relative to Gold. In
RLS detection shown here, cDNA was labeled with either fluorescein or
biotin, and detected with anti-fluorescein 60 nm Silver or anti-biotin 80 nm
Gold.
